Abstract In Indonesia, Curculigo latifolia, popularly known as lemba, is used traditional medicine for antihypertension, antidiabetic, and a food drink additive. Despite the high benefits, this plant difficult to propagate due its low seed viability. Tissue culture offers an alternative propagation, thus providing continuous source of bioactive substances. The study aimed establish efficient methods explant sterilisation, callus induction regeneration from leaf tissues multiplication. also investigated histochemically secondary metabolites produced by callus. Our results showed that sterilising agents in lowest concentrations shortest duration was most suitable getting aseptic cultures. BAP (3 mg L −1 ) IBA (5 were highest initiation, shoots, roots regeneration. histological sections fresh secretory structure form cavities surrounded layer epithelial cells idioblasts around cavities. histochemical tests identified phenolic compounds essential oils organogenic Idioblasts detected containing too. will contribute C. latifolia propagation development producing metabolites.
